Next-generation sequencing was conducted on six different tissue samples at the Genomic Lab of the David H. Murdock Research Institute(http://dhmri.org/).

For the sequencing, 6 cDNA Libraries have been prepared from the following tissues of Uca minax (commonly known as the red-jointed fiddler crab):

  • Library 1 = anterior gills RNAs
  • Library 2 = posterior gills RNAs
  • Library 3 = female + male gonads RNAs
  • Library 4 = eye stalk + muscle RNAs
  • Library 5 = 1st + 2nd Zoea stage RNAs
  • Library 6 = 3rd Zoea stage RNAs

These libraries were then combined together to generate one unified library pool for sequencing. A 100 Bp paired end read sequencing run was conducted on each individual tissue using the Illumina HiSeq2000 instrument. This yielded 12 files each containing over 7 million sequences in fastq format, each occupying over 1.8G of storage. The Sequence data quality control is:

  • 1.) Q score: > 80% of bases must have a quality score (Q) > 30 (which reflects that the probability of an incorrect base call is 1 in 1000 for an inferred base call accuracy rate of 99.9%
  • 2.) Data throughput: The data throughput QC threshold is > 100 million reads/lane.
